I'm racking my brain trying to remember which episode they introduced the Enterprise's Tactical Alert (and jokingly suggested calling it Reed Alert)... I've run a search on the terms "tactical alert" and "alert", but come up empty.

Does anyone have a better memory than me about that event?

I'm not sure if your question mark is signifying "Was the ship class named in this episode 'Neptune'?" or "Was the Neptune class the ship class named in this episode?" but the answer is yes to both.
